1️⃣ The big picture: Ancient parchment manuscripts, made from animal skins and used for centuries across Europe, the Middle East, and Africa, contain hidden genetic information that can now be studied without damaging the artifacts. Scientists at NC State University developed a technique using cytology brushes, the same type used in Pap smears, to gently collect cellular material from the surface of historic documents. Next-generation forensic sequencing tools then recover and amplify the genetic sequences found in that material. Tested on 91 manuscripts from Duke University’s Rubenstein Library, the method yielded new insights into trade routes, agricultural practices, and livestock history. The approach resolves one of the biggest barriers in this emerging field: gaining access to culturally significant artifacts whose custodians feared damage from sampling.

2️⃣ Why is this good news: For the first time, researchers can study the genetic secrets locked inside ancient manuscripts without putting these fragile, irreplaceable records at risk. This opens an entirely new and largely untapped archive of historical data, giving scientists and historians empirical evidence about how domesticated animals evolved, how livestock breeds developed, and how diseases spread across centuries. Museums, libraries, and archives that previously refused access out of conservation concerns may now feel confident allowing researchers to study their collections. Beyond history, the interdisciplinary nature of this field brings together genetics, medieval studies, veterinary science, and forensic analysis in ways that could inspire entirely new research questions. Every parchment manuscript in existence becomes a potential source of fresh knowledge, vastly expanding the raw material available to understand the ancient world.

3️⃣ What’s next: The research team is actively seeking funding to scale up their work and explore the full potential of genetic parchment analysis. Broader access to manuscript collections around the world will be key to unlocking more discoveries. Researchers also hope to build trust with institutions holding historic documents so more collections become available for study.
